Products & Film Selection 9 questions
What sealing films does PCRSeal offer?

We offer four sealing films, each designed for a specific application:

  • PCRSEAL-OA — Optical clear, pressure-sensitive adhesive (rim-seal only). Designed for qPCR / Real-Time PCR. Clean peel for downstream processing.
  • PCRSEAL-OB — Optical clear, full-surface sticky adhesive. Designed for qPCR, especially 384-well plates and high-throughput screening.
  • PCRSEAL-SF — Standard PCR sealing film for end-point PCR, sample transport, and general plate sealing.
  • PCRSEAL-ALM — Aluminum foil adhesive seal for long-term storage, compound libraries, and applications requiring a vapor barrier down to −80 °C.

We also offer the PCRSEALer — a sealing film and capping tool designed for consistent, even pressure application.

What's the difference between PCRSEAL-OA and PCRSEAL-OB?

Both are optically clear and designed for qPCR, but they differ in adhesive design:

  • PCRSEAL-OA (Model A) uses a pressure-sensitive adhesive applied only around the well rims. The adhesive doesn't contact your sample, and the film peels cleanly for downstream processing. It uses a dual adhesive system (acrylic + silicone PSA) and activates with firm pressure.
  • PCRSEAL-OB (Model B) has a full-surface sticky adhesive (acrylic). It provides maximum contact area for a strong hold, making it ideal for 384-well plates. The trade-off is that peeling is less clean than OA since adhesive covers the entire surface.

Both perform equivalently in qPCR fluorescence reads. Choose OA if you need clean peel; choose OB if you need maximum adhesion or are running 384-well plates.

Which film do I need for qPCR / Real-Time PCR?

Either PCRSEAL-OA or PCRSEAL-OB. Both are optically clear with consistent fluorescence transmission across the full excitation/emission spectrum used in SYBR Green, FAM, ROX, and multiplex assays.

Choose OA for 96-well plates or when you need clean peel for downstream processing. Choose OB for 384-well plates or when maximum adhesion is the priority.

Both films are validated on QuantStudio™, CFX96™, and LightCycler® platforms with equivalent Ct values.

Which film is best for long-term storage?

PCRSEAL-ALM (Aluminum Seal). Its aluminum foil backing provides an impervious moisture and vapor barrier, and it's rated down to −80 °C — the widest temperature range in our lineup. It's pierceable for convenient sample retrieval without removing the entire seal. Ideal for compound libraries, DMSO-containing samples, and any application requiring long-term storage integrity.

Do any of your films require a heat sealer or applicator?

No. All four PCRSeal films are adhesive-based and seal with hand pressure — no heat sealer or specialized equipment required. A sealing paddle, roller, or our PCRSEALer tool is recommended for even pressure distribution, but gloved hands work fine.

What plate formats are compatible?

All PCRSeal films are compatible with standard 96-well and 384-well plates with raised rims. Film dimensions are approximately 142 × 79.3 mm, which fits all standard SBS-format microplates.

What are the temperature ranges for each film?
  • PCRSEAL-OA: −80 °C to +120 °C
  • PCRSEAL-OB: −80 °C to +120 °C
  • PCRSEAL-SF: −80 °C to +110 °C
  • PCRSEAL-ALM: −80 °C to +120 °C
What is the shelf life and how should I store unopened film?

Store at room temperature in a cool, dry place out of direct sunlight. Films are packaged 10 per sealed bag to minimize exposure and cross-contamination. When stored properly, films maintain performance for at least 5 years from the date of manufacture.

Are the films sterile? DNase/RNase-free?

Our films are manufactured in a Class 100K cleanroom and each lot is tested for biological purity, PCR inhibitors, qPCR performance, and evaporation. They are certified PCR inhibitor-free, human DNA-free, DNase/RNase-free, ATP-free, and non-pyrogenic — tested per lot, not per process run.

Your lot-specific Certificate of Analysis documenting these results is always available on demand at pcrseal.com/coa.

How to Seal a PCR Plate 5 questions
How do I properly seal a PCR plate?
1. PREP — Ensure the plate surface is clean and dry to avoid adhesive failure. Use powder-free gloves to prevent contamination.
2. PEEL — Hold the film from one end and remove the backing in one continuous motion without touching the center sealing surface.
3. ALIGN — Position the film parallel to the edges. Secure one end first and lower it gradually to prevent curling or misalignment.
4. SEAL — Use a PCRSEALer, roller, or film paddle to apply firm pressure in one direction at a time, then perform the motion in reverse to ensure a complete bond.
5. EDGES — Firmly seal all four outside borders and corners to mitigate the "edge effect" and prevent evaporation.
6. CHECK — Visually inspect the plate for bubbles or wrinkles. A proper seal will show a clear imprint of every well.
7. BOND — Allow the plate to sit for at least 10 minutes before beginning thermal cycling to let the adhesive bond fully.
Can I peel the film after sealing?

PCRSEAL-OA is specifically designed for clean peel — since the adhesive is only around the well rims, the film lifts off cleanly for downstream processing (sequencing, cloning, etc.).

PCRSEAL-OB and PCRSEAL-SF are peelable but use full-surface adhesive, so removal is less clean.

PCRSEAL-ALM is not peelable — pierce the foil with a pipette tip to access individual wells.

What is the "edge effect" and how do I prevent it?

The edge effect refers to increased evaporation from perimeter wells due to incomplete sealing at the plate borders. To prevent it, firmly press all four outside edges and corners of the film after sealing the center. Use a sealing tool or roller along each edge to ensure full contact. This is especially important for long thermal cycling protocols.

How long should I wait before starting thermal cycling?

Allow at least 10 minutes at room temperature after sealing before beginning thermal cycling. This gives the adhesive time to bond fully, which improves seal integrity and reduces the risk of film lifting during rapid temperature transitions.

What is the PCRSEALer tool?

The PCRSEALer is a handheld sealing film and capping tool designed to apply consistent, even pressure across the plate. It's more ergonomic than a gloved finger and provides better pressure distribution than improvised tools. It also functions as an 8-strip cap remover.

Quality & Certification 5 questions
What is lot-level QC and how is it different from process certification?

Most major brands certify their manufacturing process (e.g., ISO certification of their facility). We go further — every production lot undergoes functional qPCR testing before release. This means the specific films you receive have been tested, not just the factory that made them.

Our films are manufactured in an ISO 9001:2015 and ISO 13485:2016 certified facility to PCRSeal OEM specifications.

How do I look up my Certificate of Analysis?

Visit pcrseal.com/coa, enter your lot number (printed on the box label), and your CoA will be available immediately. No waiting for paperwork, no emails to chase — available 24/7 for audits, compliance, and verification.

What does the Certificate of Analysis include?

Each CoA includes results for:

  • Biological purity testing
  • PCR inhibitor assays
  • qPCR performance validation
  • Evaporation QC

The CoA is specific to the production lot — not a generic document.

What platforms have your films been validated on?

Our optical films have been independently tested head-to-head against Applied Biosystems® and Bio-Rad® films on QuantStudio™ 384-well and CFX96™ systems with equivalent Ct values and tighter replicate consistency. They are also compatible with LightCycler® and other standard Real-Time PCR platforms.

Are PCRSeal products manufactured in an ISO-certified facility?

Yes. All PCRSeal films are manufactured in a facility certified under ISO 9001:2015 (quality management) and ISO 13485:2016 (medical device quality management) to PCRSeal OEM specifications. Films are produced in a Class 100K cleanroom.
